About Raman Manohar

I have done Ph.D. from Membrane Biology Laboratory, School of Life Sciences (SLS) under the supervision of Prof. Rajendra Prasad. Our lab is basically concerned with Candida albicans, opportunistic dimorphic pathogenic yeast, most frequently infecting the immunocompromised or patients undergoing long term chemotherapy. In recent past, development of multiple drug resistance (MDR) in C. albicans has posed a serious problem for its successful chemotherapy. Several mechanisms are known to be involved in the development of drug resistance in C. albicans. Overexpression of CDR1, an ATP Binding Cassette (ABC) transporter, is considered as one of the most important mechanism of development of drug resistance in C. albicans. Cdr1p is a membrane protein with two transmembrane domains (TMDs) and two cytosolic nucleotide-binding domains (NBDs). It is believed that the TMDs of the protein bind to a wide range of drugs/antifungals and efflux them with the help of energy derived from ATP hydrolysis by the NBDs. Thus Nucleotide Binding Domains are the hub of energy generation of drug efflux and inactivation of these NBDs abolishes the functionality of the efflux pump. Interestingly, CDR1 not only functions as a drug transporter but has several other well characterized functions attributed to it, which include transport of human steroid hormone and phospholipid translocation between the two monolayers of cell membrane.
In order to understand the mechanism of CDR1 regulation, it is necessary to carry out detailed analysis of the molecular mechanisms involved in CDR1 expression. To elucidate this aspect, I focused on dissecting the transcriptional and post-transcriptional control of CDR1 in clinical isolates of C. albicans.
